1. (a) Explain in detail principle of microscopic technique used for detection of live unstained cells. (10)
(b) Narrate the principle of flow-cytometry technique and its role in lymphocyte subset enumeration. (10)

2. Give an account on
(a) Molecular exclusion chromatography. (10)
(b) Affinity chromatography. (10)

3. (a) Explain in detail the immunoblotting technique used for diagnosis of HIV infection. (10)
(b) Give an account on kinds of ELISA and its application. (10)

4. (a) Describe in detail about measurement of radioactivity by liquid scintillation spectrophotometer. (10)
(b) Discuss the biological application of radioactive isotopes. (10)

5. Explain the principle of X-ray diffraction spectroscopy and its role in molecular structure analysis.

6. What is Proteomics? provide an account on various tools that aid proteomics.

7. Write the principle of automated DNA sequencing and its application in microbial diversity studies.

8. To verify whether a course in biotechnology improved performance, a similar test was provided to 12 participants both before and after the course. The original marks recorded in alphabetical order of the participants were 44, 40, 61, 52, 32, 44, 70, 41, 67, 72, 53, and 72. After the course , the marks were in the identical order, 53, 38, 69, 57, 46, 39,73, 48, 73, 74, 60, and 78. Using t-test obtain whether the course is useful.
